Cells of two serotypes of Escherichia coli (O26B6 and 0111B4) were counted using epifluorescence and immunofluorescence microscopy (nonavalent serum) and plate count on specific medium. Values from indirect counts were 10 fold smaller than those from both microscopic counts. In a sample of sea water from an area receiving a fecal pollution, the highest number of bacteria was obtained by epifluorescence microscopy. In that case, the number of cells counted by immunofluorescence microscopy was 10 fold smaller than the number of bacteria able to grow on the medium. The behaviour of F specific bacteriophage in depurating shellfish with reference to their use as pollution indicator organisms ArchiMer

The behaviour of sex-pilli specific (F+) bacteriophage during the depuration process was investigated using Pacific oysters (Crassostrea gigas) and edible mussels (Mytilus edulis) naturally contaminated with untreated sewage. F+ bacteriophage was eliminated considerably slower than E. coli. This effect was most pronounced in oysters which showed average T90 bacteriophage values of 62.5 hours compared with 11 hours for E. coli. F+ bacteriophage in mussels was largely confined to the digestive tract. The majority of E. coli were similarly located but, in contrast, counts were also distributed throughout the other internal tissues. The germicide properties of ultra-violet radiation in a wave length ranging from 100 to 400 mm were already discovered back in the late 19th century. These properties specific to UV-C radiation (200-280 mm) result from its action on the nucleic acids of the micro-organisms. Each micro-organism, depending on its composition, therefore presents a specific sensitivity to this radiation.

Diarréia neonatal em bovinos é um quadro complexo, normalmente responsável por altas taxas de morbidade e mortalidade. Os prejuízos são observados não somente nos gastos com a medicação, mas também na perda do animal em caso de morte e na redução do desenvolvimento e do desempenho do futuro animal adulto, comprometendo os índices de ganho de peso ou produção leiteira. A etiologia múltipla da enfermidade envolve a interação de diversas variáveis dos agentes, hospedeiro e meio ambiente. Nesta revisão são abordados os aspectos clínicos da colibacilose sanitário, a prevenção e o tratamento são também abordados.

Prevalence of enteropathogenic and enterotoxigenic Escherichia Coli in foods of animal origin in southern Brazil Ciência Rural

Prevalence of Escherichia coli in foods of animal origin from the city of Pelotas, Brazil, was determined. The ocurrence of enterotoxigenic (ETEC) and classic enteropathogenic (EPEC) serogroups among E. coli isolates was determined. E. coli was isolated from 68% of the food samples surveyed. Of 36 food samples tested, 11(30%) and 24(66%) were positive for EPEC and ETEC strains respectively. However, of 187 E. coli isolates tested, 30(16%) were EPEC compared to 75(40%) which were ETEC. The antibiotic resistance pattern revealed that the isolates were highly sensitive to all antibiotics tested.

POPULATION ANALYSIS OF SUSCEPTIBILITY TO PROPOLIS IN REFERENCE STRAINS OF Staphylococcus aureus AND Escherichia coli J. Venom. Anim. Toxins

Susceptibility to several ethanolic extracts of propolis (EEP) concentrations was tested with the population analysis technique in reference strains of Staphylococcus aureus (ATCC 25923) and Escherichia coli (ATCC 35218). The results of these tests showed that all bacteria were killed by EEP concentrations approximately equal or higher (2.0% v/v- S. aureus; 10.0% v/v - E. coli) than the respective minimal inhibitory concentrations (MIC). Regarding the susceptibility to propolis, there was a homogeneity of data with the respective time kill curves showing a clear bactericidal effect during 6 to 9 h of exposition.

In an attempt to synthesize conceptual and experimental information on the behaviour of enteric bacteria in seawater, a mathematical model has been developed. This model is based on changes in intracellular metabolisable components depending an physiological responses of the cell when subjected to nutrient starvation, salinity stress and solar radiation. Following a strategy which takes into account short- and mid-term physiological adaptation and reversal processes, the cells can develop in different states: culturable (B-1), viable and definitively nonculturable (B-2), and reversibly dormant (B-3) cells. In an ex vivo agar plate assay, we monitored the appearance of an inhibitory halo against Vibrio cholerae from the feces of Wistar and Fischer rats aged 10 to 42 days. The frequency of Wistar rats showing halo increased from 0% (10 days) to a maximum of 80.0% (29 days) and then decreased to 53.3% (42 days). A similar pattern was obtained with Fischer rats but with a lower intensity (maximum frequency of 50.0% by day 36). In a separate experiment, when Wistar rats were fed a low-protein diet for 7 days, the inhibitory halo decreased drastically. A spontaneous kanamycin-resistant Escherichia coli mutant, showing cross resistance to five other aminoglycosides and absence of the OppA protein was isolated. [3H]-dihydrostreptomycin uptake is reduced in this mutant, implying that the oligopeptide transport system is involved in accumulation of aminoglycosides, although apparently not related with aminoglycoside permeability alteration due to bacterial adaptation to osmotic changes.

Foram submetidas à pesquisa de coliformes fecais, utilizando-se a técnica do número mais provável (NMP/g), 168 amostras de variedades de queijo Minas (20 frescal, 48 canastra e 100 padronizado) coletadas em Belo Horizonte. Para a comparação de diferentes ensaios em temperatura elevada, utilizou-se o caldo EC isoladamente, e caldos EC e triptofano em paralelo. Visando à pesquisa de indol foi realizado ensaio para confirmação de produção de beta-D-glucuronidase e indol em caldo fluorocult lauril sulfato. Fermentation of sweet whey by recombinant Escherichia coli K011 BJM

The production of ethanol from sweet whey using the recombinant Escherichia coli KO11, in batch fermentation, was tested. The maximum ethanol yield was reached after 96h, representing only 38% of the theoretical yield. The supplementation of whey with components of LB broth increased the maximum yield to 96% in 72h. The addition of 0.5% yeast extract to whey resulted in maximum yield of 74% at 36h and it increased to over 100% when yeast extract and trace metals solution (Fe++, Mn++ and Zn++) were added.

Motilidade espermática e integridade acrossomal em doses de sêmen suíno refrigeradas e inoculadas com Esecherichia coli e Staphylococcus aureus Ciência Rural

Foram utilizados 12 ejaculados, coletados de maneira asséptica, distribuídos em sete tratamentos, sendo um grupo controle e os demais inoculados com três diferentes concentrações de S. aureus ou E. coli (5 x 10(5), 5 x 10(6) e 5 x 10(7) UFC/ml). Durante 96 horas, foram avaliados a motilidade espermática, o percentual de acrossomas intactos (NAR), o pH e o número de unidades formadoras de colônia (UFC/ml) das bactérias inoculadas. O desenvolvimento bacteriano foi decrescente ao longo das 96 horas de armazenamento. Com exceção do tratamento com a inoculação de 5 x 10(7) UFC de E. coli/ml, não foi observado efeito significativo das bactérias sobre a motilidade espermática (p>0,05). The sanitary classification of harvesting areas for bivalve mollusks in France is based on the level of Escherichia Coli contamination detected in shellfish meat, as defined in EC Directive 91/492 EEC. However, outbreaks of gastroenteritis or hepatitis after consumption of shellfish meeting current bacteriological standards suggest that E. coli is a poor indicator of viral contamination. Characterization of a highly thermostable alkaline phosphatase from the euryarchaeon Pyrococcus abyssi ArchiMer

This work reports the first isolation and characterization of an alkaline phosphatase (AP) from a hyperthermophilic archaeon. An AP gene from Pyrococcus abyssi, a euryarchaeon isolated from a deep-sea hydrothermal vent, was cloned and the enzyme expressed in Escherichia coli. Analysis of the sequence showed conservation of the active site and structural elements of the E. coli AP. The recombinant AP was purified and characterized. Monomeric and homodimeric active forms were detected, with a monomer molecular mass of 54 kDa. Apparent optimum pH and temperature were estimated at 11.0 and 70 degreesC, respectively. Thus far, P. abyssi AP has been demonstrated to be the most thermostable AP, with half-lives at 100 and 105 degreesC of 18 and 5 h, respectively....
